Summary. The aim of the research was to study the effect of different nutrient media, as well as their individual components, on growth and development of apical meristem of local and new grape varieties. The work was carried out in 2010-2013. The study subject was 10 grape varieties of different applications (4 table varieties, 2 universal and 4 technical ones). Five mediums were tested for cultivation of apical meristem. Further cultivation was carried out on nutrient media studied early. Development of apical meristem of test varieties at introduction in vitro was different. The fraction of viable meristem was on average 35.5 %. The lowest value of this indicator was identified on the standard solid medium, it was 21.6 %. For other media it was similar: MS 1/Krymsk – 44.4 %, MS 1 liq. – 44.1 %, and it was little bit higher on medium MS 1-2 mod. – 45.2 %. On this medium the mitotic activity of apex was higher, that contributed to the formation of a powerful conglomerate of buds and side shoots. During cultivation of meristem the varietal specificity to the medium MS 1 liq. strongly appeared, the survival rate ranged from 0 to 100 %. Among studied genotypes varieties Diamant, Shokoladny and Livadiisky Cherny were the most successful in induction of meristem. Their viability was 55.3, 48.4 and 46.2 %, correspondingly. The lowest value of this index, 12.6 %, was observed for variety Pervenets Magaracha. Varietal differences were apparent both at the stage of proliferation and elongation. The multiplication factor of the separate variety was similar at both stages. It was the highest for technical varieties Muscat Golodrigi (1:7.2 and 1:5.6) and Saperavi (1:6.8 and 1:7.5). The value of multiplication factor of table varieties was much lower. These special features should be considered when planning the production of planting stock.
