Abstract. This paper studies the polymorphism of kappa-casein (CSN 3) gene in dairy cattle herds and the influence of genotypes on the milk productivity of cows of different lines. The work was performed in 2014–2015 in two dairy herds (n = 264 and n = 238, the Republic of Tatarstan) on full-growth cows of the Black-and-White breed of Vis Beck Idial 101341, Montwick Chieftain 95679, and Reflection Sovering 198998 lines. In the studies we used the method of polymerase chain reaction with artificial restriction fragment length polymorphism (A-RFLP) analysis. Milk productivity for 305 days during 3rd and later lactations was determined individually according to the results of monthly control milkings. The frequency of the occurrence of the desired CSN3 BB genotype in the cows of V.B. Ideal line was the lowest amounting to 1.1–2.1 %. The occurrence frequency of allele B was 0.16–0.22. The occurrence frequency of CSN3 BB genotype and allele B in the R. Sovering line was 5.5–8.1% and 0.24–0.27, in the M. Chieftain line it amounted to 5.7–7.4% and 0.25–0.28 respectively. The highest performance in terms of milk yield, mass content of fat and amount of milk fat showed the cows of V. B. Idial and M. Chieftain lines, carriers of allele A. The cows of M. Chiftain line with CSN3 AA genotype were much more productive in terms of in milk yield – by 317–495 kg, but were inferior in mass fraction of protein in milk – by 0.05–0.15%. In the R. Sovering line, the best in terms of milk productivity were the carriers of the allele B. Compared to different lines in the CSN3 AA group, the cows of V.B. Idial line had the advantage: in terms of milk yield – by 102–174 kg of milk, in terms of milk fat – by 4.6–6.2 kg, in terms of milk protein – by 2.1–6.4 kg. In CSN3 АВ and CSN3 ВВ groups the best performance showed the cows of R. Sovering line. The CSN3 AB group significantly exceeded the M. Chiftain line in terms of the milk yield – by 335 kg of milk, in terms of the milk fat yield – by 10.5 kg, and in terms of milk protein – by 11.7 kg.
