Biotechnological methods for reproduction of decorative Hydrangea L. varieties

Abstract. The studies were conducted at the Main Botanical Garden of the Russian Academy of Sciences in 2017–2020. The studies aimed to determine the characteristics of clonal micropropagation of varieties of three species of the genus hydrangea: Hydrangea arborescens (Sterilis); H. macrophylla (Bodensee, Forever and Ever Blue); and H. paniculata (Candlelight, Magical Candle, Polar Bear, Praecox, and Wim’s Red). At the stage of introduction into in vitro culture, lateral buds and metameres of young shoots were used as initial explants. At the sterilization stage, the optimal duration of exposure to calcium hypochlorite was in the range from 7 minutes to 10 minutes (at 1-minute intervals) with the sequential use of solutions of the systemic fungicide benomyl (2%), ethanol (70%), calcium hypochlorite (7%), and mercuric chloride (0.1%). At the stage of micropropagation, we selected the best concentration of cytokinin 6-BAP out of the three studied concentrations: 0.5 mg/L, 1.0 mg/L, and 2.0 mg/L. The control was the MS culture medium without growth regulators. The most effective sterilization was provided by the use of 7% calcium hypochlorite for 8 minutes. The yield of viable explants, in this case, reached 68–79%. The share of the genotype influence on the realization of the morphogenetic potential of the studied varieties was 42%; the share of the influence of the hormonal composition of the nutrient medium was 22%. The highest multiplication factor in H. arborescens and H. macrophylla was observed when they were cultivated in the MS medium containing 6-BAP at the concentration of 1.0 mg/L; for H. paniculata Candlelight and Wim’s Red this indicator was 2.0 mg/L; for Magical Candle and Polar Bear this indicator was 1.0 mg/L. In terms of regenerative capacity, the highest morphogenetic potential was characteristic of H. paniculata varieties (8.1); the morphogenetic potential of H. macrophylla and H. arborescens varieties was significantly lower – 4.0 and 2.9, respectively.
