Genetic structure of the Pechora population of semifine-wool sheep by alleles of STR-loci

Abstract. To assess the genetic diversity and structure of semifine-wool sheep of the Pechora population, 59 ewes of the gene pool flock were genotyped for 12 polymorphic STR-loci. The entire genotyped livestock was divided into 2 subpopulations according to the origin. The first subpopulation (1.1) included crossbreeds obtained from crossing animals of the Pechora breed group with the Romney Marsh and Kuibyshev breeds. The second one (1.2) included crossbreeds derived from crossing individuals of the northern native, Pechora breed group and the Romney Marsh breed. A total of 94 alleles were identified for 12 microsatellite loci in both subpopulations, of which 84 alleles were found in the first subpopulation, and 80 alleles in the second. The highest allelic diversity was found for the INRA005 and INRA023 loci, the lowest for MAF214, and the excess of heterozygotes for the MAF065 and INRA063 loci. In subpopulation 1.1 positive FIS values were recorded at five loci, and in subpopulation 1.2 at two loci. In subpopulation 1.2 the excess of heterozygotes was higher than in 1.1. Avarage FIS indices for 12 microsatellite loci in each of the subpopulations were negative. Differences between subpopulations in FIS values, total and effective number of alleles per locus, level of expected and observed heterozygosity were not significant. The genetic differentiation between subpopulations, estimated using FST, was 0.015 while the Nei genetic distance (DN) was 0.089. 12 private alleles were identified in subpopulation 1.1, and 8 were found in subpopulation 1.2. Considering the phenotypic, breed, and genealogical differences between the subpopulations, the identified private alleles in 1.2 seem to characterize the relict allele pool of the northern native sheep. The private alleles of subpopulation 1.1 were the allele pool of the Kuibyshev breed.
