POLYMORPHISM OF -824 A/G TUMOR NECROSIS FACTOR ALPHA GENE AND INDICATORS OF CATTLE REPRODUCTION

Summary. Tumor necrosis factor alpha (TNF-alpha) gene encodes the protein which is a multifunctional cytokine, and along with the functions of the immune response it is involved in the morphogenesis and fetation, plays the important role in the formation of a mammary gland, stimulates the formation of prostaglandins. The purpose of our study was to investigate the possible association between -824 A/G polymorphism in the promoter of TNF-alpha gene and reproductive indices (age at the first insemination, service period, calving interval, interlactation period, pregnancy) of Red Steppe cattle. Polymorphism of TNF-alpha gene was studied using polymerase chain reaction followed by restriction fragment length analysis. For the analysis DNA was extracted by the standard method of protease treatment followed by extraction with phenol-chloroform from the blood of 148 cows of Red Steppe breed from agricultural cooperative stud farm “Stepnoy” in Altai Territory. Reproduction indices were determined on the basis of zootechnical calculation. In the studied group the frequencies of genotypes G/G, G/A and A/A for TNF-alpha gene was 56, 38 and 6 % respectively. The frequency of the rare allele A was 25 %, of allele G – 75 %. The distribution of genotype frequencies corresponded to the Hardy-Weinberg equilibrium distribution. The age of animals with genotype G/G at first insemination was 2 months less compared to G/A genotype (p < 0.05). The duration of pregnancy in the animals with G/G genotype was higher than that in G/A genotype by 6.6 days (p < 0.05). No significant associations were found for other reproductive performance (service period, calving interval and interlactation period) between cows with different genotypes.
