EVALUATION OF EFFICIENCY FOR IN VITRO EMBRYO PRODUCTION USING SEX-SORTED SPERM IN CATTLE

Summary. Using sex – preselected in vitro embryos for animal breeding can significantly increase the profitability of dairy and beef cattle production. However there is a need to improve some stages of this technology. We examined the embryonic development of cow oocytes in vitro depending on the sperm preparation and presence of heparin in fertilization medium. Oocyte-cumulus complexes were isolated from ovaries of butchered cows and heifers and cultured in groups of 25-40 complexes in the medium TC199 for 24 h. Frozen unsorted and sex-sorted sperm samples were thawed and separated by either a swim-up or Percoll gradient procedure. Mature oocytes were co-incubated during 18 h with prepared sperm in the medium Fert-Talp supplemented with 10 or 2 microg/ml (only for sex-sorted sperm) of heparine, after then them were replaced in embryos medium and cultured until the blastocyst state. It was revealed the lower fraction of cleavage and blastocyst rate with using sex-sorted sperm compared with those derived with unsorted sperm (57.2 ± 3.4 and 22.3 ± 2.3% vs 75.9 ± 2.3 and 37.8 ± 2.1% respectively). The result of fertilization in both cases depended on the sperm preparation method. Swim-up treatment was better for unsorted sperm and Percoll gradient procedure was better for sex-sorted sperm. The decrease in the heparine concentration in the fertilization medium reduced the fraction of oocyte cleavage, inseminated by the sex-selected sperm, but did not influence their ability to further embryonic development. It was recommended to use Percoll gradient procedure for preparation of sex-sorted sperm to obtain the fraction of active spermatozoa.
