INVESTIGATION OF QUANTITATIVE AND QUALITATIVE CHARACTERISTICS OF SEMEN OF TRANSGENIC ROOSTERS

Summary. The aim of the study was to evaluate the influence of integration of the recombinant DNA on the qualitative and quantitative characteristics of semen of transgenic roosters. The studies were carried out at the vivarium of L.K. Ernst Institute of Animal Husbandry in 2015. Transgenic roosters and their non-transgenic analogues of the cross Shaver Brown were the objects of the study. The transgenic chickens were produced by using a lentiviral vector, which contained the reporter gene GFP (green fluorescent protein). We examined the following quantitative and qualitative characteristics of semen of transgenic (n = 8) and non-transgenic (control) roosters (n = 8): the volume of ejaculate, sperm motility and their concentration; length, width, perimeter, head area, tail length and the total spermatozoa length; length, width, perimeter, area and acrosome preservation. The evaluation of these characteristics was carried out according to the standard techniques using software NIS-Elements BR 4.30. The morphometric analysis of spermatozoa and assessment of the acrosome status in the transgenic and non-transgenic roosters showed the increase in sperm head area by 15% (Р is more than 0.95) with the decrease in acrosome area by 7% in transgenic roosters. At the same time, deviations in acrosome preservation in transgenic roosters compared to the control were not revealed. It was registered the tendency to the reduction of sperm quantitative characteristics among transgenic roosters: the volume of ejaculate, concentration and motility of spermatozoa compared to the control were lower by 20, 5 and 16%, correspondingly. This may indicate a certain negative influence of transgene integration on the functional capacity of germ cells in test transgenic roosters.
