Summary. The investigations were carried out on the base of laboratory of biotechnology and molecular biology of the All-Russia Research Institute of Rice Breeding under conditions of laboratory experiment in 2013-2016. The goal of the study was to develop a methodology system for evaluation of genetic purity of seeds F1 of Brassica oleracea L., based on polymorphism of microsatellite DNA-markers. Genotypes of headed cabbage used by the department of vegetables and potato breeding of the All-Russia Research Institute of Rice Breeding for creation of hybrids F1 were used as a material for the research. Nineteen co-dominant microsatellite (SSR) markers were included in the work. DNA extraction was performed by CTAB-method. From the test set of microsatellite markers the greatest polymorphism between the parental forms was foe loci Na12-F12 and Na12-A02. Noticeable differences in specific alleles were observed foe loci Ra2-E12, Ni2B02, SSR 10, 6 and 7. For all other studied SSR-markers the differences in the DNA-profile between the parental lines of Brassica oleracea L. were not found. For 19 loci in 5 samples of cabbage 28 allelic states were found (on average 3 alleles per a locus). The highest number of alleles (6-8) for a selected set of genotypes was detected for loci Na12-A02, Na12-F12, localized on different chromosomes of the SSR marker, the smallest (1-2) – for the loci Na10-D09, Ra2-E12 and Ni3-G04B. When testing 19 SSR markers three most informative and localized on different chromosomes SSR markers – Na12-A02, Na12-F12 and Ra2-E12 showed the highest efficiency in detecting interspecific polymorphism of collection samples of headed cabbage from the vegetable breeding department. They can be recommended for use in further work with PCR-method for evaluating hybridity of commercial seeds of hybrids F1 of headed cabbage.
