Summary. Ex situ preservation of collections of cultivated species of potato requires the development of effective methods for eradication of viruses from the collection material. This article presents the comparison of two methods of virus eradication, used to remove one of the most common and harmful virus – PLRV (potato leafroll virus) from potato plants. The work was carried out in the biotechnology department of N.I.Vavilov Institute of Plant Genetic Resources (VIR) in 2014-2016. The material for the study consisted of plants of 15 samples of aborigine Chilean cultivars of potato (Solanum tuberosum L.), which were reproduced for a long time in the field collection of VIR, and also derived from them in vitro plants, in which the presence of PLRV was confirmed by enzyme immunoassay and RT-PCR. The complex therapy included three successive cycles, each of which consisted of chemotherapy and thermotherapy (cultivation of microplants for 4 weeks at 35 degrees on a medium containing ribavirin in the concentration of 30 mg/l). Cryotherapy was performed using the droplet-vitrification protocol; the duration of the exposure of plant material in liquid nitrogen was 1 hour. The presence or absence of PLRV in microplants before and after the therapy was tested by RT-PCR. The results indicate higher (p is less than 0.05) efficiency of cryotherapy compared with the complex therapy. Percentage of virus-free from PLRV potato microplants in the case of complex therapy was 63.2%, and after application of cryotherapy - 92.9%.
