Summary. We examined DNA samples from cows of Simmental breed of the local breeding (n = 86) from SKhPK “Ust’-Aldan”, Ust-Aldan district; Kholmogory breed (n = 38) from OOO “Kladovaya Olekmy”, Olekminsk district; Simmental breed of Austrian breeding (n = 75) and Kalmyk breed (n = 12) from OOO “Agrofirma Nemyugyu”, Khangalassky district of the Sakha (Yakutia) Republic. The experiments were carried out in the Laboratory of breeding and genetics at the premises of Yakutsk State Agricultural Academy. It was revealed two alleles of kappa-casein gene: CSN3A and CSN3B, and three genotypes: CSN3AA, CSN3AB and CSN3BB, with greater frequency of CSN3A allele, which varied depending on the breed from 0.50 to 0.79. The highest frequency of CSN3A allele (0.79) was registered in cows of Austrian Simmental breed. In cattle of Kholmogory and Kalmyk breeds and Simmental breed of local and Austrian breeding the frequency of LGBA allele varied from 0.17 to 0.47. It was revealed two alleles of prolactin gene: PRLA and PRLB, and three genotypes: PRLAA, PRLAB and PRLBB. The frequency of PRLA allele (0.79-0.84) significantly prevails over the frequency of PRLB allele (0.16-0.21) in all investigated cattle breeds (p is less 0.001). Simmental breed of the local breeding possesses the highest polymorphy in the three studied genes (1.691) and the highest degree of heterozygosity (0.400). In the studied cattle breeds there is an excess of heterozygotes, as indicated by the calculated inbreeding coefficient for the three genes, which has a negative value in Kalmyk (-0.154), Austrian Simmental breed (-0.122) and the local Simmental breed (-0.065) and slightly positive value in Kholmogory breed (0.035).
