Influence of Different Carbon and Nitrogen Sources on the Production of Xylanases by Bipolaris sorokiniana

Abstract. The effect of carbon and nitrogen sources in different concentrations on the production of xylanases by Bipolaris sorokiniana Bp1 was studied. The strain was cultivated in liquid nutrition medium by the pour plate method at 30 C for 5 days: yeast extract – 5.0 g/L, Na2HPO4*2H2O – 10.0 g/L, KCl – 0.5 g/L and MgSO4*7H2O – 0.15 g/L. The hydrolase activity of xylanases was determined by staining the reducing sugars with dinitrosalicylic acid, 1% birch xylan was a substrate. As a source of carbon for B. sorokiniana Bp1 there were used glucose, maltose, xylose, cellulose, starch, different agricultural and industrial wastes: wheat straw, wheat bran, barley straw, corn cobs, sawdust. Ammonium nitrate, ammonium sulfate, sodium nitrate, potassium nitrate, peptone, urea and yeast extract were used as sources of nitrogen. The maximum activity of xylanases (43.69 U/mL on the 5th day) was obtained on the medium with 4% wheat straw. An increase in the enzymatic activity of hydrolase was observed when carbon sources such as xylan, barley straw, corn cobs were added (18.78, 12.84 and 17.26 U/mL, respectively). The highest repression of xylanase production was caused by raffinose, glucose, xylose and maltose (0.06, 0.09, 0.19 and 0.27 U/mL, respectively). The addition of 0.5% sodium nitrate to the culture medium resulted in a 4-fold increase in xylanase activity compared to the control. The addition of ammonium chloride, nitrate and sulfate had a negligible effect on the production of hydrolytic enzymes in culture liquid of B.sorokiniana Bp1 (12.11, 12.60 and 13.78 U/mL, respectively).
