Study of the Influence of Mycomycetes’ Metabolites of Fusarium Genus on Callus Oat Culture

Abstract. In 2017–2018 we conducted an in vitro study on the effect of the toxic fungi metabolites Fusarium poae (Peck) Wollenweber, in Lewis (1913) and Fusarium equiseti (Corda) Saccardo (1886) on the processes of morphogenesis and regeneration of plants in the culture of spring oats young embryos. The nutrient media were prepared according to the recipe of Murashige and Skoog. The culture filtrates containing toxic metabolites were isolated on the basis of isolates from the collection of oats root rot pathogens. The culture filtrates were used as selective agents on the medium for callus proliferation. For the fungi of F. poae species we used 30% and 40% (of the medium volume) concentrations; for the fungi of F. eqiseti species we used 30%, 40% and 50% concentrations. The reaction of the callus cultures in response to the introduction of F. poae and F. eqiseti metabolites into the medium was assessed by three parameters: the change in the callus volume, organogenesis manifestation, formation of full-fledged regenerant plants. Two thousand five hundred seventy-eight young embryos of eight bare-grained and glumaceous oats variety were introduced into the culture. The action of micromycetes metabolites at the stage of elongation was the distress of the callus volume (by 49.4–71.0% for F. poae and 37.6–66.1% for F. eqiseti), organogenesis manifestations (by 30.7–44.0% and 31.4–41.3%, respectively), plant regeneration (by 29.9–32.3% and 21.5–26.2%, respectively). The negative after-effects of toxins were noted at the stage of plant regeneration (by 19.4–33.0% and by 7.62–15.2%, respectively). Under in vitro conditions F. poae culture filtrates exerted a stronger inhibitory effect on morphogenesis stages than F. eqiseti filtrates. The varieties were different in their response to micromycetes metabolites in terms of organogenesis and regeneration on the medium of callus proliferation with the 40% culture filtrates of F. poae. Golets, Tubinsky and Zolotoy Pochatok varieties were referred to the most susceptible to the action of two species of Fusarium – regeneration on the media containing culture filtrates decreased to zero or was 2–3 times less than the average regeneration for the conditions. Sayan and Selma varieties were greatly inhibited by the culture filtrates of F. poae only reducing regeneration from 12.5% to 0%.
