Accelerated Development of High-Quality Analogues of Durum Wheat Genotypes Adapted to the Middle Volga Conditions Using Genomic Selection Methods

Abstract. The purpose of the studies was to develop methods for the accelerated introduction of genes encoding high-quality gluten into an adapted gene pool with the prospect of developing competitive varieties of spring durum wheat in Russia. The work was performed in the Middle Volga region in 2016–2019. The methodology for the use of marker-associated (MAS) and genomic selection (GS) was tested. MAS technology was used to identify genetic systems encoded gluten quality. The biochemical marker Glu–A1 (7+8) and the molecular markers of the microsatellite groups of SSR — Single sequence Repeat (Barc148) and SNP – Single Nucleotide Polymorphism (BM140362) were used. To identify genetic systems of adaptability in recombinant plants we used GS technology with 100 molecular markers of SSR. As a result, we bred Taganrog variety, which has been undergoing State variety testing since 2018, and the analogues of SP-3-2 selection line, which will be tested in the Russian variety testing system in 2021. According to the results of testing in 12 variety test plots in the Middle Volga region (Samara, Penza, and Ulyanovsk regions, Republic of Mordovia and Tatarstan), Taganrog exceeded the average standard in the yield by 2.3 kg/ha, or by more than 11.0%. In variety test plots, grain yield of Taganrog varied from 1.1 t/ha to 4.7 t/ha. In 9 out of 12 test plots, it exceeded the standards for some indicators. The high adaptability of two high-quality analogues of SP-3-2 breeding line was established in field tests at the Samara Research Institute of Agriculture in 2019. Their yield was 1.24 t/ha and 1.3 t/ha; the yield of Bezenchukskaya 210 standard was 1.29 t/ha; the yield of the recurrent parent (SP-3-2 breeding line) was 1.21 t/ha. The developed varieties are promising for cultivation in the steppe areas of the Volga region and the Russian Federation.
