Abstract. Transvaginal ultrasound-assisted follicular puncture – Ovum-Pick-Up (OPU) – is used for intravital production of cattle ova. The purpose of our studies was to determine the effect of the multiplicity of OPU sessions on the number and quality of oocytes obtained from cows of Simmental breed. The work was performed on sexually mature heifers of Simmental breed aged from 16 to 19 months (n = 4) with a natural sexual cycle. Follicle aspiration was performed once (1/w) or twice (2/w) per week. The same donor animals were used for each regimen. During OPU sessions in the 1/w mode, 228 follicles were aspirated; in the 2/w mode, 224 follicles were aspirated. From them 140 oocyte-cumulus complexes were extracted, or (61.4 ± 5.9)% and (62.1 ± 6.3)%, respectively. On average, 4.4 oocytes per session were received from each donor. When performing the procedure in 1/w mode, the proportion of good quality OPU oocytes with normal morphology was (53.6 ± 3.0)% of the total number recovered; in 2/w mode, it was (65.7 ± 4.0)%, which is significantly higher – 1.23 times. In 24 hours of maturation, the fractions of oocytes at the stage of MII meiosis ((74.7 ± 2.4)% and (73.3 ± 5.6)%, respectively) and oocytes with the signs of apoptotic changes ((9.8 ± 3.3)% and (8.0 ± 3.1)%, respectively) did not significantly differ among groups. The multiplicity of OPU sessions did not affect the degree of embryo fragmentation (63.5% on average) and their development up to the blastocyst stage (16.7% on average). The OPU procedure twice a week made it possible to obtain 2.46 times more embryos in the blastocyst stage through in vitro technology from heifers of Simmental breed compared to the case it was performed once a week.
