Using Driver & Kuniyuki Walnut medium for micropropagation of red raspberry varieties

Abstract. The purpose of the studies was to evaluate the prospects of using DKW medium (Driver & Kuniyuki Walnut medium) compared to MS medium (Murasige & Scoog medium) for the clonal micropropagation of red raspberries of normal and remontant types of fruiting. The object of the study was raspberry varieties of common (Avrora, Akvarel, Barnaulskaya, Zatonskaya, Meteor, Troyana) and remontant (Atlant, Bryanskoe Divo, Gerakl, Oranzhevoe Chudo, Pingvin) types of fruiting. The work was performed in three stages. At the first stage, we compared the mineral media DKW and MS supplemented with six concentrations of growth regulators (GR): 6-benzyl amino purine (BAP) in the range of 0.7–7.0 uM and alpha-indolebutyric acid (IBA) in the range of 0.014–0.14 uM in a ratio of 5:1. The second stage involved the optimization of the passage duration (45 and 60 days) on the DKW medium supplemented with three concentrations of the growth regulators BAP/IBA – 1.5/0.3–3.5/0.7 uM. At the third stage, the optimal concentration of GR was determined for groups of raspberry varieties of normal and remontant types of fruiting in DKW medium supplemented with four concentrations of BAP / IBA in the range of 1.5 / 0.3 to 4.5 / 0.9 uM with a passage duration of 60 days. Reproduction factor, shoot length, and the number of leaves on the shoot of microplants on DKW medium was 1.5–1.8 times higher than on MS medium. The shoots on the DKW medium were also superior in terms of quality indicators (shape and colour of the leaves). The change in the duration of the passage from 45 to 60 days contributed to an increase in the length of microshoots by two times and the number of leaves by 1.3 times. Optimal concentrations of BAP and IBA with a passage duration of 60 days were 2.5–3.5 uM and 0.5–0.7 uM, respectively. The remontant varieties in DKW medium exceeded the ordinary varieties in terms of the reproduction factor by 30%; in terms of shoot length in the option with 4.5 uM of BAP and 0.9 uM of IBA, they exceeded them 1.7 times; in terms of the number of leaves on the shoot, they exceeded them by 10%.
