Abstract. Sonographic-assisted transvaginal follicle puncture (Ovum Pick-Up, OPU) is the most common method for multiple collection of bovine oocytes from living females. The effectiveness of OPU depends on many factors, including breed and individual characteristics of donors, technical and technological parameters of OPU, etc. The study aimed to determine the technical parameters (such as aspiration needle and vacuum pressure) that increase the effectiveness of OPU in Simmental heifers (n = 5). The follicles were aspirated using a system for conducting OPU in cattle (Minitube, Germany). The interval between OPU sessions was seven days. Two different types of aspiration needles (18G and 20G) were tested at a vacuum pressure level of 80 mmHg and two different vacuum pressure levels (80 mmHg and 90 mmHg) using an 18G needle. The criteria for evaluating the effectiveness of OPU were the recovery rate of cumulus-oocyte complexes (COCs) and the number of COCs suitable for producing embryos in vitro. When using a needle with a larger diameter (18G), compared with a needle with a smaller diameter (20G), the recovery rate of COCs increased from 40.91% to 52.17% (by 11.26%, p = 0.2239). This increased the total number of COCs from one donor per session (from 1.80 to 2.40) and the number of eligible COCs (from 1.60 to 2.00). Increasing the vacuum pressure from 80 mmHg to 90 mmHg when using an 18G aspiration needle led to an increase in the recovery rate of COCs from 48.35% to 60.68% and, as a result, to obtaining more COCs from one donor per session (3.43 vs. 2.20). At the same time, the quality of the resulting COCs remained almost unchanged - the proportion of suitable COCs was 72.73% and 74.65%, respectively. In general, in the case of intravital sonographic-assisted oocyte retrieval in Simmental heifers, the best results in terms of recovery rate and the number of obtained cumulus-oocyte complexes were registered in the case of using aspiration 18G needle and at a vacuum pressure of 90 mmHg. Such technical parameters can be recommended for routine OPU in Simmental donor heifers.
