Screening of new strains of granulosis virus against the greater wax moth Galleria mellonella

Abstract. The studies aimed to determine the possibility of using Galleria mellonella as a test object for screening strains of codling moth granulosis virus from the Bioresource Collection of the Federal Scientific Centre for Biological Plant Protection in 2020-2021. Inoculation on an artificial nutrient medium was carried out by the surface method. During the development of viral biomass, C. pomonella caterpillars were kept at an air temperature of 26 C and a humidity of 70%, while G. mellonella caterpillars were kept at 30 C and 70%, respectively. During the research we searched for granules in the tissues of insects, which were formed as a result of the viruses’ vital activity, and conducted a comparative analysis of their size. The diameter of the granules in the body of C. pomonella was 0.62-2.19 um and they had a typical rounded shape. Granules in the body of G. mellonella were found in smaller quantities. Their diameter was 0.38-1.34 um (0.2 um less than the standard), and they had an irregular shape. All studied insects had similar manifestations of the virosis pathological process (rarefaction of internal tissues and organs, skin discoloration). ‘Madex Twin, SK’ served as a biological standard during the evaluation of seasonal activity. Conditions for keeping insects were the following: air temperature was 30 C, humidity was 70%. Data obtained in summer and winter periods were compared. In summer, the biological efficiency of the studied strains was 2 times higher than in winter. The most promising strains of granulosis virus were identified according to the criterion of biological effectiveness against the greater wax moth: BZR GV 5, BZR GV 10, BZR GV 14, BZR GV L-7.
