APPLICATION OF DNA DIAGNOSTICS IN SANITATION PROGRAMS FOR ERADICATION OF BOVINE LEUCOSIS

Summary. The characteristics of different methods (RIA, ELISA, PCR) used for diagnostics of bovine leukemia virus (BLV) was done. The advantages and disadvantages of their applications in programs for leucosis eradications were discussed. The effectiveness of PCR method as the additional confirmation test in frame of systematic sanitary procedure was shown. The analysis of epizootic situation regarding bovine leucosis in the different regions of the Russian Federation was performed. The aim of the present study was the development and application of the test-system for diagnostics of BLV using real-time PCR. The research results showed the high analytical sensitivity of the developed test-system for identification of animals carrying the BLV (1.12*103 copies/ml). The analysis of the control blood panel and 192 blood samples of cows using serological methods (RIA, ELISA) and the real-time PCR method (developed test-system) demonstrated the complete coincidence of research results. Thus the diagnostic specificity and sensitivity of developed test-system was 100 per cent. The practical application of developed test-system allowed identifying the virus carriers among calves in the age up to 6 months: 6 BLV carriers were detected (15.0 per cent). The effectiveness of the test-system application for the analysis of the pulled blood samples was shown: the analysis of 21 pulled probes allowed correctly identifying of three BLV carriers among 100 individual samples that made it possible essentially decrease the expenses and labor-intensiveness of the process. The developed test-system for BLV diagnostics on the base of real-time PCR method could be rated as the effective tool to increase the effectiveness of sanitary procedure for bovine leucosis.
