IMMUNOLOGICAL AND MORPHOLOGICAL ESTIMATION OF IMMUNOMODULATING ACTION OF KIM-M2 IN GUINEA PIGS INFECTED WITH MYCOBACTERIAS

Summary. The investigations were conducted to study the effects of complex microbial immunomodulator KIM-M2 on guinea pigs infected with a virulent strain of Mycobacterium bovis and atypical Mycobacterium scrofulaceum. For the experiment we selected 30 adult male guinea pigs and formed 6 groups from them. Five intact guinea pigs (group I) served as the control. We injected subcutaneously M. scrofulaceum at the dose of 30 mg to the animals of group II-IV (n = 5), after that we injected KIM-M2 subcutaneously at the dose of 500 microgram/ml of protein to the animals from III and IV groups in 14 and 28 days, correspondingly. Guinea pigs from V and VI groups (n = 5) were injected with virulent culture of M. bovis strain 14 subcutaneously at the dose of 0.0001 mg/ml, in addition, animals from VI group (n = 5) were administered with KIM-M2 subcutaneously at the dose of 0.5 ml (500 micrograms of protein) 30 days before it. At the first stage of the investigation we quantified the content of immune cells in the blood of guinea pigs from all groups; on the second step we estimated the structural and functional state of the liver of guinea pigs immunized with KIM-M2 after infection with M. bovis and M. scrofulaceum. The introduction of specific immunomodulator contributed to the formation of higher level of antituberculous protection, which was evidenced by the significant increase (p < 0.05) in the number of T-, B-lymphocytes and lymphocytes with killer activity in individuals from III, IV and VI groups. The analysis of the structural and functional state of the liver of guinea pigs immunized KIM-M2 after infection with M. bovis and M. scrofulaceum, carried out at the next stage of the investigation, showed that the drug contributed to the reduction of the tinctorial, dystrophic and destructive changes of hepatocytes, reduced the number and volume of granulomas in liver, enhanced the compensatory-regenerative processes in the parenchyma of liver.
