IDENTIFICATION OF GENES ENCODING ECONOMICALLY VALUABLE CHARACTERS OF SUNFLOWER BY MOLECULAR SCREENING

Summary. We carried out the molecular screening of 96 lines from genetic collection, which included 92 samples with different ability for fertility restoration in crosses with lines with CMS, and 4 sterile lines based on CMS PET1 and RIG0. The aim of the screening was to identify the sources of genes encoding pollen fertility restoration and resistance to rust. With the use of STS marker orfH522 and genealogical analysis the presence of sterile cytoplasm of PET1 type was confirmed for 78 fertile lines. This indicated the presence of Rf1 karyogene in their genotypes. The SCAR markers HRG01, HRG02, marker STS115, as well as SSR-markers ORS224, ORS511, ORS799 localized in the linkage group 13 at different distance from the locus Rf1, were used for identification of this gene. Markers HRG01, HRG02 and STS115 were characterized by the highest diagnostic value for detection of sources of Rf1 gene. This enables to recommend them for efficient selection of carriers of Rf1 gene from segregating hybrid populations. A scheme for using of HRG01, HRG02 and STS115 markers for genetic purity testing of hybrid parental lines and hybridity level is suggested. With the use of SCAR markers SCO04, SCX20 and SCT06 the lines were differentiated depending on the presence or absence of rust resistance genes R1 and Radv. The sterile lines based on CMS PET1 lacked R1 and Radv genes. Both genes were identified in 34 lines, only one gene (R1) was found in 34 lines, and in five lines only Radv gene was registered with the absence of R1. With the use of five linked markers gene Rf1 and rust resistance genes R1 and Radv were found in lines RIL80, RIL130, VIR130, VIR234, VIR249, VIR361, VIR369, VIR378 and VIR740.
