CRYOPRESERVATION OF VEGETATIVE BUDS OF SWEET AND SOUR CHERRY USING CRYOPROTECTORS

Summary. The aim of the study was to develop a convenient and reliable method for cryopreservation of vegetative buds of sour cherry (Prunus subg. Cerasus (MILL.) A. GRAY, 1856) and sweet cherry (Prunus avium (L.) L., 1755) with the use of cryoprotectants. It was studied the effect of two cryoprotectants – glycerol and sucrose in different concentrations - on the viability of vegetative buds of these cultures after storage in vapour phase nitrogen.The research material consisted of sweet and sour cherry cultivars grown in the collection orchards in different parts of Russia: Southern (Krymsk experiment breeding station of VIR) and North-Western (Pavlovsk experiment station of VIR) regions. Freezing was carried out by two-step method. The samples were stored in liquid nitrogen vapor at a temperature of -183…-185 deg C, then they were thawed gradually with the interval of 1 deg C/min up to 0 deg C and with subsequent washing off from the cryoprotectors at 18…20 deg C for 1…2 min. When the buds were defrosted, their viability varied depending on the cultivar and the region of growing, but it was high enough at all the samples. It was 54.2…63.2 % for buds of sour cherry and 36.0… 56.3 % for buds of sweet cherry. It was found a tendency for higher viability of buds after cryopreservation with 40 % sucrose solution. The proposed conservation technique when buds undergoes two-step freezing with different cryoprotectants is less expensive than in vivo maintenance of the collection in orchards. In addition, cryopreservation of buds did not require a long preparatory period, in contrast to cryopreservation of cuttings. In general, two-step cryopreservation of buds of sour and sweet cherries using 40 % sucrose solution as a cryoprotectant can be used for conservation of genetic resources of these cultures.
