DEVELOPMENT OF TEST SYSTEM FOR DIAGNOSTICS OF CATTLE FERTILITY HAPLOTYPE HH3 ASSOCIATED WITH EARLY EMBRYONIC MORTALITY

Summary. Increase in the level of homozygosity of cattle breeds contributes to the growing negative impact of LoF-mutations (LoF – loss of function). The aim of this work was to develop a molecular genetic test system for diagnostics of LoF mutation in SMC2 gene, known as fertility haplotype HH3, which is associated with early embryonic mortality. At the first stage of the research, we created the series of reference samples with known genotypes for HH3 using direct DNA sequence analysis by pyrosequencing. At the second stage we evaluated the applicability of the test system by genotyping reference samples. At the third stage we determined the distribution of HH3 in the Russian population of Holstein and holsteinized Black-and-White cattle. Proposed test system is based on the use of single tube allele-specific PCR method (STAS PCR). Because of genotyping by pyrosequencing we created a series of reference samples (n = 60), including 5 samples with TC genotype (latent carriers for HH3) and 55 samples with TT genotype (non-carriers for HH3). The analysis of reference samples using the developed test system showed full concordance of results of genotyping. A study of 353 cows and 632 bulls revealed the presence of 49 carriers of HH3, including 35 cows and 14 bulls, which corresponds to the frequencies of 9.9 and 2.2%, respectively. Taking in into account the relatively high distribution of HH3, we recommend using the developed test system for screening cattle to control the spread and to eliminate this genetic disorder among Russian breeding cattle population.
