THE USE OF DNA-TECHNOLOGIES TO DEVELOP RICE VARIETIES RESISTANT TO BLAST

Summary. The development of basic methodologies of marker assisted breeding of rice resistant to Pyricularia oryzae Cav. (Magnaporthe grisea (Herbert) Barr Yaegashi & Udagawa (anamorphic Pyricularia grisea, rice blast disease)), as well as development of modern genetic resources resistant to this disease is one of the priority tasks in the world rice breeding. Using modern technologies of molecular DNA-marking it was realized the introduction and pyramiding of blast resistance genes Pi-ta, Pi-b, Pi-1, Pi-2, Pi-33 into domestic rice varieties for providing them long-term resistance to the disease. During our work with the help of marker control of donor alleles we obtained valuable breeding material with pyramided genes of resistance to the blast disease (Pi- 2, Pi-b, Pi-ta), (Pi-2 , Pi-b, Pi-33). According to the results of phytopathological test it is resistant to a mixture of pathogen isolates, selected in Krasnodar Krai. The transfer of dominant alleles of each gene in the progeny was controlled by codominant closely linked SSR- markers established in All-Russian Research Institute of Rice Breeding. To improve the economic efficiency of marker breeding, we proposed multiplex PCR system, enabling the identification of two genes (Pi-1 + Pi-2, Pi-33 + Pi-ta, Pi-ta + Pi-b) of resistance to M. grisea in the genomic DNA of hybrid plants during one PCR-reaction. The use of such reaction will significantly reduce the cost of consumables and time to perform the analysis.
