REGENERATIVE PECULIARITIES OF PRIMARY EXPLANTS OF PHLOX PANICULATA L. IN VITRO

Summary. The regenerative peculiarities of primary explants of summer phlox (Phlox paniculata L.) at the stages of introduction in vitro and subsequent propagation were studied. Inflorescence fragments (flower with a pedicle of 10-15 mm in length), apical and axile shoots were used as explants. Three cultivars of summer phlox from the collection of the M.A. Lisavenko Research Institute of Horticulture of Siberia were the objects of the investigations. Nutritional media were prepared according to Murashige and Skoog (MS) with an addition of 30 g/l sucrose and growth regulators: alpha-naphthyl acetic acid (NAA), beta-indolyl-3-butyric acid (IBA), 6-benzylaminopurin (BAP), thidiazuron (TDZ), kinetin (Kn). Step treatment of shoots with 70% alcohol and 0.1% solution of corrosive sublimate gave a possibility to yield 60-80% of sterile and viable explants. Change of sublimate by “Domestos” (active ingredient is sodium hypochlorite) increased sterility and viability of Areole shoots up to 95%, for other cultivars results were comparable. At application of two-stage sterilization by 70% alcohol and 0.1% solution of HgCl2, the output of sterile floral explants reached 80-100%. Direct regeneration of shoots from apical and axile meristem (1-2 pieces on an explant) without callus formation on the base part of microcuttings was observed in MS nutritional medium with 0.5 mcM Kn + 0.5 mcM IBA. In MS media with application of 10.0-20.0 mcM BAP + 1.0 mcM NAA + 0.1 mcM IBA, or 15.0 mcM TDZ+ 1.0 mcM NAA + 0.1 mcM IBA) it was registered the direct gemmogenesis and rhizogenesis from flower bud tissues. The average number of shoots on an explant depended on a genotype and reached 7.6 for the Butonnik variety, 5.2 – Areole variety and 5.1 – Dragon variety. Embryos developed from prominences in the lower part of explants under the action of BAP 20 mcM in combination with NAA 1 mcM and IBA 0.1 mcM.
