CHARACTERISTICS OF QUALITY PARAMETERS OF SEMEN DERIVED FROM HYBRID RAMS WITH DIFFERENT BLOOD RATIO FOR ARGALI

Abstract. Saving of genetic resources in vitro in the form of cryobanks is the priority method for conservation of genetic resources. The aim of this work was to evaluate the quality characteristics of freshly derived and frozen-thawed semen of rams of different genetic origin. The investigation was carried out in the L.K. Ernst All-Russian Research Institute of Animal Husbandry in 2014-2016. As the material for our study we used the semen, which was derived from rams, having the different blood ratio for Romanov breed and argali: F1 (n = 1, 50 % of argali blood; it was obtained by surgical insemination of Romanov sheep with frozen-thawed semen of argali), backcross BC1 (n = 5, 25 % of the argali blood; obtained by crossing Romanov sheep with F1 ram), backcross BC2 (n = 5, 12.5 % of argali blood; obtained by crossing Romanov sheep with BC1 rams). The sperm of the Romanov ram (ROM) was used as a control. All rams had the identical feeding and maintenance conditions. For freshly derived semen, the ratio of spermatozoa with straight forward movement (classes A+B) in groups ROM, F1, BC1 and BC2 was, respectively, (93.0 ± 1.7), (92.7 ± 4.0), (90.6 ± 2.4) and (88.9 ± 2.1) %. Cryopreservation led to a significant decline in the proportion of spermatozoa of A+B classes in all groups, respectively, to (42.2 ± 1.5), (52.3 ± 8.4), (43.6 ± 3.9) and (42.0 ± 2.8) %. We did not observed the significant differences in the ratio of spermatozoa with intact acrosome (the values for freshly derived semen ranged from (89.9 ± 2.9) % in BC1 to (96.5 ± 6.7) % in F1; for the frozen-thawed semen – from (85.2 ± 2.9) % in BC2 to (90.3 ± 8.7) % in ROM) and in the ratio of spermatozoa with damaged DNA (the values for freshly derived semen ranged from (8.6 ± 2.8) % in F1 to (12.7 ± 1.7) % in ROM; for frozen-thawed semen – from (11.6 ± 3.9) % in ROM to (14.8 ± 2.5) % in BC1). Our data will be used to develop quality requirements for semen, derived from rams with different blood ratio for argali to create cryobanks of generative material.
