DORMANT PERIOD AND SAFETY OF POTATO MICROTUBERS IN VITRO UNDER VARIOUS TEMPERATURE REGIMES OF STORAGE

Abstract. We compared the duration of a dormant period of in vitro potato microtubers, depending on temperature conditions of the storage process. To produce microtubers, plastic containers were used with two variants of liquid nutrient medium with a different concentration of sucrose. Regeneration of explants occurred in the medium with 3 percent of sucrose, 16-hours photoperiod and the temperature 21–22 Celsius degrees. To induce tuber formation, the medium was replaced with a medium with 8% concentration of sucrose, followed by placing the containers in a short day (8/16 h day/night) conditions at 18-20 Celsius degrees during the day and 10-12 Celsius degrees at night. Formation and ripening of microtubers occurred in the continuous dark with the temperature of (18 ± 2) Celsius degrees. After harvesting, the material was stored under two temperature regimes: 4-6 Celsius degrees and 6-8 Celsius degrees. The end of the dormant period was determined visually when a sprout 2-3 mm long appeared on the tuber. The temperature regime of storage has a significant effect on the duration of the rest period of microtubers (81.48 %). The share of the influence of varietal features was 10.18 % of the total variation. The duration of the rest period does not correlate with the early ripeness. The duration of the rest period of microtubers was 3.5-4.5 months at the storage temperature from +6 to +8 Celsius degrees and 6-7 months at the temperature from +4 to +6 Celsius degrees. The duration of the period from the beginning of germination to the end of the rest was 20-40 and 20-50 days, respectively. The results of the assessment of the safety of microtubers after 12 months of storage showed that at the temperature regime from +4 to +6 Celsius degrees it was 80-87 %.
