Abstract. Genetic certification and identification of pigs is an urgent task in selection and breeding work. Microsatellites, so-called STR-markers, are used for these purposes. The aim of the current study was genetic identification and certification of Kemerovo breed of pigs with the use of STR-analysis, as well as an estimation of its genetic diversity and differentiation degree in comparison with other local and commercial breeds. The studies were carried out in 2017 on 79 animals of three local breeds: Kemerovo (KEM, n = 31), Berkshire (BER, n = 29), Large Black (LB, n = 19) and 121 animals of three commercial breeds: Large White (LW, n = 64), Landrace (L, n = 21) and Duroc (DU, n = 36). Tissue samples (ear notches) of pigs, available in the collection of the bank of genetic material of domestic and wild animals and poultry, L.K. Ernst All-Russian Institute of Animal Husbandry, were used for the study. Genomic DNA was extracted using Nextech column (Germany) according to manufacturer's recommendations. Polymorphism of 10 STR-markers (SW24, S0155, S0355, S0386, SW72, SW951, S0101, SW240, SW857and SO005) was determined by own procedures on ABI 3130xl DNA analyzer («Applied Biosystems», USA). Statistical analysis was performed in MS Excel 2007 with GenAIEx v. 6.5 plugin. Genetic structure of the studied breeds was assessed through clustering in STRUCTURE 2.3.4 software. Kemerovo breed has the lowest values of the observed degree of heterozygosity in comparison with Landrace, Duroc, and Large White breeds: 0.554 ± 0.054 vs. 0.635 ± 0.082, 0.567 ± 0.092, 0.762 ± 0.053, respectively. However, it exceeds the local breeds (Large Black and Berkshire) by this index: 0.542 ± 0.079 and 0.499 ± 0.072. Kemerovo pigs are characterized by the maximum values of the mean (Na) and effective (Ne) numbers of alleles, yielding only to Large White breed (Na(KEM) = 5.000 ± 0.645, Na(LW) = 5.556 ± 0.852, Ne(KEM) = 3.295 ± 0.384, Ne(LW) = 3.305 ± 0.446). The results of the genetic structure analysis showed that all studied breeds form their own cluster with a high degree of genetic consolidation, which indicates the uniqueness of their allele pool.
