Abstract. We studied various schemes of obtaining anti-species monospecific serums based on the use of killed Brucella cultures in combination with an oil adjuvant MONTANIDE(TM) ISA 61 VG (SEPPIS, France), required for the bacteriological diagnosis of brucellosis. The investigations were carried out at the premises of the All-Russian Research Institute of Brucellosis and Tuberculosis of Animals in 2015–2016. For the production of anti-melitensis serum two groups of rabbits were formed from healthy animals, three heads in each group. Animals from the group I were administered once intravenously by 1ml of a live culture of B. melitensis 16M (200 million CFU/ml). Animals from the group II were administered once subcutaneously by 1ml of the inactivated culture of B. melitensis 16M (200 million CFU/ml) in a mixture with adjuvant (40 and 60%, respectively). To produce anti-abortus serum, four groups in three rabbits were formed. Group I rabbits were injected intravenously with 1 ml of the mixture from a suspension of inactivated and live B. abortus 19 cultures with a concentration of 4*10E9 CFU/ml (1: 1); II, III and IV groups – once subcutaneously into the throatiness area of 1 ml of inactivated B. abortus 19 culture (200, 100 and 300 million CFU/ml, respectively) in admixture with the adjuvant. Anti-melitensis and anti-abortus serums were obtained by adsorption of the Brucella suspension of heterologous species. Optimal schemes were tested on animals of the second groups. In this case, the serums retained their activity (the agglutination reaction at a dilution of not less than 1: 160) for at least 6 months after receiving from the blood taken in 21, 28, 35 and 60 days after hyperimmunization. Serums obtained from rabbits from the first groups with their bleeding in 5–7 days after immunization lost diagnostic activity during 28–35 days. The use of the adjuvant reduces the labor intensity of the process, increases its anti-epidemic safety, as well as the volume of serum with high activity, due to the possibility of taking blood at least 4 times from one animal.
