TAXONOMIC AFFILIATION OF PHYTOPLASMA, IDENTIFIED IN POTATO IN THE RUSSIAN FEDERATION

Abstract. From 2006 to 2016, 1,347 potato leaf samples, 713 individual Hemiptera insects from Moscow region and 48 insects from Samara region were analyzed. The aim of the studies was to determine the taxonomy affiliation of phytoplasma causing potato diseases and infecting insect-vectors in various economic regions of the Russian Federation. PCR was used to detect phytoplasma DNA in the samples, RFLP-analysis with restriction endonucleases АluI, MseI, HhaI, HpaII, and TaqI was applied to identify taxonomic affiliation of phytoplasmas. In the Central economic region, the samples were collected annually, in the Northern, North-Western, Central Black Earth, North Caucasian, Volga, Urals and West Siberian regions they were collected in some years. The percentage of phytoplasma infection was on average 27%. In the Central economic region 194 samples, or 21.2%, from 923 samples were infected. Thirty-four percent of them were infected with the phytoplasma belonging to purple top wilt of potato group (16SrI), 30% – with the phytoplasma of X-disease group (16SrIII), 18% – with the phytoplasma from the clover proliferation group (16SrVI), and 17% – with the phytoplasma from Stolbur group (16SrXII), less than 1% were the samples, infected with the phytoplasma of Peanut witches’-broom group (16SrII). In addition to the Central region, 16SrII phytoplasma group was also found in the Northern region, and 16SrXII group phytoplasma was found in samples from all surveyed regions. Phytoplasma from group 16SrIII was in the samples from the Central, Volga, North Caucasian, Ural and Western-Siberian regions. Group 16SrI phytoplasma occurred in the Northern, Central Black Earth and North-Caucasian regions, and phytoplasma of group 16SrVI was in the West-Siberian region. In Moscow region, among nineteen identified species of the order Hemiptera, 10 species were carriers of phytoplasma. Euscelis incisus Krs. and Macrosteles laevis Rib. prevailed, the other species of carriers were presented by single individuals. Among nine species of the order Hemiptera collected in Samara region, phytoplasmas typical for potato were discovered in Hyalesthes obsoletus Sign. and Pentastiridius leporinus L. (16SrXII phytoplasma group), and in Psammotettix striatus L. (16SrIII phytoplasma group). The prevalence of phytoplasma in the region depends on the insect vectors living there.
