METAGENOMIC ANALYSIS OF THE MICROBIOTA CHANGE OF COW RUMEN WITH THE USE OF EXPERIMENTAL FEED CONCENTRATE

Abstract. The purpose of the research was a comparative analysis of changes in the rumen microbiota of dairy cows when experimental feed concentrate was included in diets. The studies were performed in the Republic of Tatarstan in 2017–2018 on 80 dairy cows of Kholmogorskaya breed of the Tatarstan type. The animals were divided into four groups of 20 animals each. Cows from the control group (I) were fed by the basic diet. Animals from experimental groups II, III, and IV were fed by 100, 150 and 200 g/head of the experimental feed concentrate in addition to the basic diet. The additive included a complex of enzymes and probiotic microorganisms (5%), L-carnitine (5%) and sapropel (90%). Rumen microflora analysis was performed using sequencing of the 16S rRNA gene on the IlluminaMiSec platform. The obtained metagenomic data was analyzed with the help of the QIIME pipeline using the Greengenes v.13.8 database and the RDP Classifier. In animals from groups III and IV, it was revealed the highest content of microorganisms of genera of Fibrobacter (0.0110–0.0162% and 0.0070–0.0160%), Ruminococcus (0.0275– 0.0493% and 0.0250–0.0437%); Anaeroplasma (0.0330–0.0580% and 0.0220–0.0610%); Ruminobacter (0.0003–0.0036 and 0.0022–0.0064%). In animals from group I, the share of these microorganisms was, respectively, 0.0035–0.0122, 0.0150–0.0200, 0.0080–0.0430, 0.0002–0.0064%. The Shannon index in animals from group I was 8.21, II – 7.50, III – 8.80, IV – 8.18. Perhaps, the most balanced representation of the rumen microbiota in cows of group III was facilitated by the optimally selected dose of the experimental feed concentrate.
