Use of DNA Markers to Examine the Source Breeding Material of Potato

Abstract. In 2018, in the selection and technology center for potato breeding, Ural Research Institute of Agriculture, varieties and interspecific hybrids of potato were screened using seven DNA markers. Based on published data and our own experiments, we selected PCR markers to identify genes of vertical resistance in potato genotypes against the most common diseases and pests. Eighty-nine samples were analyzed, including 27 varieties and 62 interspecific hybrids. In the collection it was found a high occurrence of genes from S. andigenum – TG-689 marker of H1 gene of resistance to a golden potato nematode, which was isolated from 92% of the samples. The presence of Gro 1-4 gene from the wild species S. spegazzinii was noted in 60% of the samples analyzed. Marker RxSP of resistance to X-virus from S. andigenum was identified in 32% of the samples. Rpi-sto 1-890 marker of late blight resistance from S. stoloniferum was represented in the collection with a high frequency – 84%, and a marker of Rpi-blb 1 gene from S. bulbocastanum was extremely rare; it was isolated from only one sample of SM 503-55, in which late blight resistance was confirmed by phenotypic studies. Samples SM 503-55, 14-45-4, Alaska, Getsbi, Gornyak, Irbitsky, Memfis, Mishka, Nicse, Terra, Evalution, 12-47-10, 14-4-1, which were characterized by high saturation with markers of disease and pest resistance genes, are recommended as the most valuable breeding material.
