Creation of Genetic Passports of Apple Rootstock Forms on the Basis of Microsatellite DNA Polymorphism

Abstract. Polymorphism of microsatellite loci for the genetic identification of apple rootstock forms M9, B9, 54-118, 62-396 was analyzed. A diagnostic set from 10 SSR markers (CH01c06, CH01f03b, CH02c02b, CH02g04, CH03a04, CH03d12, CH04c07, CH04h02, CH05e04, and CH05g08) was formed. The proposed set allowed reliable identification of apple rootstock forms. The work was carried out in 2017–2018. The replication of the experiments was 3-fold. Plant material of apple rootstocks B9, 62-396, 54-118 was taken in the genetic collection of the I.V. Michurin Federal Scientific Center, M9 apple rootstock was received from Sady Stavropolia. Fragment analysis of the obtained PCR products was performed using Applied Biosystems 3730 DNA Analyzer automated sequencer, which allows determining the size of SSR alleles with an accuracy of one nucleotide. The obtained chromatograms were processed in the GeneMarker V2.6.3 software environment. In the studied apple rootstocks, the analyzed set of microsatellites was represented by 39 alleles, the size of which was in the range of 94–202 bp. Among the 39 alleles, 37 were polymorphic, i.e. could detect differences between samples. The number of alleles per marker ranged from 2 (marker CH05e04) to 5 (markers CH02g04, CH04c07). The average level of polymorphism of the studied SSR loci was 94.8%, the level of heterozygosity was 82.0%. For the M9 stock, the share of unique alleles was 41.2% with an average heterozygosity level of 82.3%; for B9 the values of these parameters were 10.5% and 94.7%; for 54-118 – 22.2% and 88.9%; for 62-396 – 15.8% and 94.7%, respectively. The low fraction of unique SSR alleles in B9, 54-118 and 62-396 rootstocks was due to the fact that forms 54-118 and 62-396 were obtained using the B9 stock as the parent form. Based on the data obtained, genetic passports of apple rootstock forms were formed.
