Genetic variability of lace bugs (Hemiptera: Tingidae) in the Krasnodar Territory

Abstract. The studies were carried out in 2019–2021 in territories of invasive distribution of lace bugs (Hеmiptera: Tingidae) in the Krasnodar Territory. The work aimed to study the process of species divergence based on the analysis of the variability of the molecular genetic structure and the genetic similarity of the geographical populations of lace bugs in the south of Russia. PCR analysis of adventive (Corythucha arcuata Say, Corythucha ciliata Say) and native (Stephanitis pyri F.) Tingidae species was performed for RAPD markers (random amplified polymorphic DNA). The study object were geographical samples of insects (n = 20) from the lace bug habitats in the Krasnodar Territory, collected in sample trees of English oak (Quercus robur L.), oriental plane (Platanus orientalis L.), common pear (Pyrus communis L.). Operon Technology (OP) primers with the standard nucleotide sequence OPA07 (GAAACGGGTG), OPA09 (GGGTAACGCC), and OPA18 (AGGTGACCGT) were used in PCR. Statistical data processing was performed using Popgene software, version 1.31. The share of intrapopulation variation of three studied species of lace bugs accounted for 93.2% (oak), 90.2% (plane) and 96.4% (pear). The value of the coefficient Gst characterised the variability between populations, which determines the differentiation between them. The interpopulation variability in adventive species was 6.8% (C. arcuata) and 9.8% (C. ciliata), while in the native species S. pyri it was 3.6%. A higher rate of divergence (Vdiv. = 0.0050 c.u./year) was noted in the species of oak lace bug recently penetrated the south of Russia (2015).
